Is there a method to allow internally developed app on Supervised iPad to allow access to Microphone and Camera access on the device without someone interacting with the device? I didn't see anything in configuration profile but I'm not sure on the development side if they can do anything since this is internally developed app. Since this is a kiosk so, I don't want customers or employees messing around with the settings to get it working.

No, Apple does not allow it. I am not a programmer either, but from what I have seen, any app accessing the camera or microphone will trigger a prompt automatically. It can't be bypassed as that would be a huge security hole.
