Can update check in normally, but a large number of Configuration profiles pending and failing

I don't think anyone can do anything with this little information. you have to show the CP, maybe post the error message as well

Are ANY configuration profiles deploying? Or are some working and some aren't? If it's the former, then you likely need to work with whichever team or person manages your network (firewalls, proxy, etc.) APNs is very touchy, and needs to have the proper URLs and ports open for communication on your network, or your Macs simply won't be able to install the profiles. Refer to Apple's documentation on this - https://support.apple.com/en-us/HT210060

If some of your profiles are working, and some aren't, then it's likely some other issue. Also, are the ones that are pending or failing happening across all devices, or just some of them? These are all key things to understand to be able to troubleshoot this.
