I'm going to say this probably cannot be done programmatically.

Entourage stores its signatures in its Database file and that file can
only be accessed and modified by Entourage itself. The file is in a
proprietary format and not something that can be edited using Defaults or
an XML editor.

The only way to manipulate Entourage programmatically is with AppleScript
or Automator, however, their dictionaries contains no entries for
signatures. You can't create them, delete them or modify them using
AppleScript.

You may be able to accomplish this using AppleScript with System Events
commands to call out menus, click buttons and type text, but that's clunky
and I haven't tested that it will even work with Entourage. Doing
something like this could be confusing to users if you don't tell them
what's about to happen.

You may need to rely on your mail sever to append a common signature to
all outbound mail messages or simply provide each user a set of
instructions for adding the new signature themselves.
