It is my assumption that VPP updates only really work when the app is closed. As Slack is something that is usually always running and never closed, I'm not sure how VPP could update it, itself.
Regardless of whether a user is admin (many of our users are not) , can the Slack app update itself without having to go through VPP and without any additional scripting? I believe it has a built in auto updater?

Slack has a different release channel for the App Store and for the installer that they distribute. Slack will not auto-update itself unless the app is closed if you are distributing it via VPP/App Store.
You will want to distribute the app by any other means if you want to let it update itself. Having done both types of deployments, the auto update feature is infinitely nicer via the native app and not via VPP.
